The Duke Human Vaccine Institute and the Center for HIV/AIDS Vaccine Immunology & Immunogen Design (CHAVI-ID), providing national and international leadership in the fight against major infectious diseases, is currently recruiting for a Postdoctoral Associate positions. The Duke Human Vaccine Institute (DHVI) is an interdisciplinary, interdepartmental institute dedicated to the study of basic and translational science required to understand host-pathogen interactions that can be translated to vaccines against human diseases. DHVI comprises a team of highly interactive investigators that have expertise in mucosal and systemic virology, molecular biology, microbiology and animal models.

 

We are seeking a highly motivated recent graduate to join our teaminvolved in the assessment of Vaccine Induced HIV-1 Env Antibody Responses. The most efficacious HIV-1 vaccine to date elicited non- neutralizing antibody responses whose titers inversely correlated with infection risk. Additionally, the non- neutralizing antibody effector function, antibody-dependent cellular cytotoxicity (ADCC) inversely correlated with infection risk. Therefore, vaccines that elicit high titers of ADCC-mediating antibodies are of interest. The Postdoctoral Associate will work as partof a team to design novel mRNA vaccines to elicit protective HIV-1 antibody responses. The candidate will compare immunogens produced from the mRNA platform to other vaccine platforms such as recombinant protein. The Postdoctoral Associate will work independently to isolate and characterize HIV-1 antibodies from vaccinated guinea pigs and nonhuman primates to discern whether vaccine- induced antibodies target known ADCC epitopes. Additionally, the Postdoctoral Associate will work independently to performimmunoassays characterizing the functions of the antibodies. These assays will include ELISA, luminex, phagocytosis, ADCC and complement fixation assays for serum polyclonal antibodies as well as monoclonal antibodies isolated from vaccinated animals. Under the guidance of the Principal investigator, the candidate will be responsible for designing and executing experiments, interpreting the data, presenting the results at internal and external meetings, and drafting scientific publications.

 

More specifically, the Postdoctoral Associate with perform the following activities:

• Transient transfection of mRNA and DNA envelope constructs to evaluate expression.

• Antigenic characterization of HIV-1 envelope by flow cytometry and ELISA

• Design, expression,and purification of HIV-1 envelopes for use in immunoassays.

• Fluorescence-activated cell sorting of single B cells from vaccinated animals.

• PCR amplification of antibody genes.

• Expression of recombinant HIV-1 antibodies by transient transfection.

• Illumina MiSeq next generation sequencing of antibody genes.

• Antibody sequences analysis. • Immunoassays to include (luminex, ELISA, and Western Blots).

• Present scientific results at meetings (internal and external) and publish results in peer-reviewed journals.

• Participation in training opportunities for bioethics, laboratory management, and scientific writing.

• Maintain accurate laboratory documentation of experiments, including raw experimental data and laboratory notebooks.

 

Education

The position requires a MD or PhD in biological sciences, chemistry, or a related discipline 

Experience

This position requires a strong record of publications in peer-reviewed journals. Ideal candidates will possess excellent oral and written communication skills with the ability to work in a highly collaborative team environment.
